Fucosyltransferase
A fucosyltransferase is an enzyme that transfers an L-fucose sugar from a GDP-fucose (guanosine diphosphate-fucose) donor substrate to an acceptor substrate. The acceptor substrate can be another sugar such as the transfer of a fucose to a core GlcNAc (N-acetylglucosamine) sugar as in the case of N-linked glycosylation, or to a protein, as in the case of ''O''-linked glycosylation produced by ''O''-fucosyltransferase. There are various fucosyltransferases in mammals, the vast majority of which, are located in the Golgi apparatus. The ''O''-fucosyltransferases have recently been shown to localize to the endoplasmic reticulum (ER). Some of the proteins in this group are responsible for the molecular basis of the blood group antigens, surface markers on the outside of the red blood cell membrane. Most of these markers are proteins, but some are carbohydrates attached to lipids or proteins eid M.E., Lomas-Francis C. The Blood Group Antigen FactsBook Academic Press, London / San D ...

FUT3
Galactoside 3(4)-L-fucosyltransferase is an enzyme that in humans is encoded by the ''FUT3'' gene. Function The Lewis histo-blood group system comprises a set of fucosylated glycosphingolipids that are synthesized by exocrine epithelial cells and circulate in body fluids. The glycosphingolipids function in embryogenesis, tissue differentiation, tumor metastasis, inflammation, and bacterial adhesion. They are secondarily absorbed to red blood cells giving rise to their Lewis phenotype. This gene is a member of the fucosyltransferase family, which catalyzes the addition of fucose to precursor polysaccharides in the last step of Lewis antigen biosynthesis. It encodes an enzyme with alpha(1,3)-fucosyltransferase and alpha(1,4)-fucosyltransferase activities. Mutations in this gene are responsible for the majority of Lewis antigen-negative phenotypes. Multiple alternatively spliced variants, encoding the same protein, have been found for this gene. See also * Cluster of differe ...

FUT6
Alpha-(1,3)-fucosyltransferase is an enzyme that in humans is encoded by the ''FUT6'' gene. The alpha-1,3-fucosyltransferases constitute a large family of glycosyltransferases with a high degree of homology. The enzymes of this family comprise 3 main activity patterns called myeloid, plasma, and Lewis, based on their capacity to transfer alpha-L-fucose to distinct oligosaccharide acceptors, their sensitivity to N-ethylmaleimide inhibition, their cation An ion () is an atom or molecule with a net electrical charge. GDP-fucose Protein O-fucosyltransferase 1
GDP-fucose protein ''O''-fucosyltransferase 1 also known as peptide-O-fucosyltransferase 1 (O-FucT-1) is an enzyme that in humans is encoded by the POFUT1 gene. POFUT-1 belongs to the O-Fuc family of proteins all which is involved in the transferring of o-fucose from GDP-β-L-fucose to substrates. POFUT-1 responsible for adding fucose sugars in ''O'' linkage to serine or threonine residues between the second and third conserved cysteines in EGF-like repeats on the Notch protein. The protein is an inverting glycosyltransferase, which means that the enzyme uses GDP-β-L-fucose as a donor substrate and transfers the fucose in ''O'' linkage to the protein producing fucose-α-''O''-serine/threonine. When the gene for POFUT1 is knocked out, or the expression is decreased to very low levels, all Notch signaling is destroyed, which means that fucose on Notch is essential for Notch function. Why this is the case is not yet well understood. Almost all glycosyltransferases reside in the G ...

Lewis Antigen System
The Lewis antigen system is a human blood group system. It is based upon two genes on chromosome 19: FUT3, or Lewis gene; and FUT2, or Secretor gene. Both genes are expressed in glandular epithelia. FUT2 has a dominant allele which codes for an enzyme (designated Se) and a recessive allele which does not produce a functional enzyme (designated se). Similarly, FUT3 has a functional dominant allele (Le) and a non-functional recessive allele (le). The proteins produced by the FUT2 and FUT3 genes modify type I oligosaccharide chains to create Lewis antigens. These oligosaccharide chains are similar to the type II chains of the ABO blood system, with a single bond in a different position. The link between the Lewis blood group and secretion of the ABO blood group antigens was possibly the first example of multiple effects of a human gene: the same enzyme (fucosyltransferase2) which converts the Le-a antigen to Le-b is also responsible for the presence of soluble A, B and H antigens in ...
