Galactoside 3(4)-L-fucosyltransferase is an enzyme that, in humans, is encoded by the ''FUT3'' gene. Function The Lewis histo-blood group system comprises a set of fucosylated glycosphingolipids synthesized by exocrine epithelial cells and circulating in body fluids. These glycosphingolipids play essential roles in embryogenesis, tissue differentiation, tumor metastasis, inflammation, and bacterial adhesion. They are secondarily absorbed onto red blood cells, giving rise to the Lewis phenotype. This gene is a member of the fucosyltransferase family, which catalyzes the addition of fucose to precursor polysaccharides during the final step of Lewis antigen biosynthesis. It encodes an enzyme with both alpha(1,3)-fucosyltransferase and alpha(1,4)-fucosyltransferase activities. Mutations in this gene are responsible for most Lewis antigen-negative phenotypes. Lewis Blood-group System
The Lewis antigen system is a human blood group system. It is based upon two genes on chromosome 19: FUT3, or Lewis gene; and FUT2, or Secretor gene. Both genes are expressed in glandular epithelia. FUT2 has a dominant allele which codes for an enzyme (designated Se) and a recessive allele which does not produce a functional enzyme (designated se). Similarly, FUT3 has a functional dominant allele (Le) and a non-functional recessive allele (le). The proteins produced by the FUT2 and FUT3 genes modify type I oligosaccharide chains to create Lewis antigens. These oligosaccharide chains are similar to the type II chains of the ABO blood system, with a single bond in a different position. The cluster of differentiation (also known as cluster of designation or classification determinant and often abbreviated as CD) is a protocol used for the identification and investigation of cell surface molecules providing targets for immunophenotyping of cells. In terms of physiology, CD molecules can act in numerous ways, often acting as receptors or ligands important to the cell.
