Meselson–Stahl Experiment
The Meselson–Stahl experiment is an experiment by Matthew Meselson and Franklin Stahl in 1958 which supported Watson and Crick's hypothesis that DNA replication was semiconservative. In semiconservative replication, when the double-stranded DNA helix is replicated, each of the two new double-stranded DNA helices consisted of one strand from the original helix and one newly synthesized. Matthew Meselson
Matthew Stanley Meselson (born May 24, 1930) is a geneticist and molecular biologist currently at Harvard University, known for his demonstration, with Franklin Stahl, of semi-conservative DNA replication. After completing his Ph.D. under Linus Pauling at the California Institute of Technology, Meselson became a Professor at Harvard University in 1960, where he has remained today as Professor of the Natural Sciences. In the famous Meselson–Stahl experiment of 1958 he and Frank Stahl demonstrated through nitrogen isotope labeling that DNA is replicated semi-conservatively. In addition, Meselson, François Jacob, and Sydney Brenner discovered the existence of messenger RNA in 1961. Max Ludwig Henning Delbrück (; September 4, 1906 – March 9, 1981) was a German–American biophysicist who participated in launching the molecular biology research program in the late 1930s. Caesium chloride or cesium chloride is the inorganic compound with the formula Caesium, CsChloride, Cl. This colorless salt is an important source of caesium ions in a variety of niche applications. Buoyant density centrifugation (also isopycnic centrifugation or equilibrium density-gradient centrifugation) uses the concept of buoyancy to separate molecules in solution by their differences in density. Implementation Historically a cesium chloride (CsCl) solution was often used, but more commonly used density gradients are sucrose or Percoll. This application requires a solution with high density and yet relatively low viscosity, and CsCl suits it because of its high solubility in water, high density owing to the large mass of Cs, as well as low viscosity and high stability of CsCl solutions.
