Genetics Of Ageing
Genetics of aging is generally concerned with life extension associated with genetic alterations, rather than with Progeroid syndromes, accelerated aging diseases leading to reduction in lifespan. The first mutation found to increase longevity in an animal was the ''age-1'' gene in ''Caenorhabditis elegans''. Michael Klass discovered that lifespan of ''C. elegans'' could be altered by mutations, but Klass believed that the effect was due to reduced food consumption (calorie restriction). Thomas Johnson later showed that life extension of up to 65% was due to the mutation itself rather than due to calorie restriction, and he named the gene ''age-1'' in the expectation that other genes that control aging would be found. The ''age-1'' gene encodes the catalytic subunit of Class I PI 3-kinases, class-I phosphatidylinositol 3-kinase (PI3K). Ultraviolet radiation, also known as simply UV, is electromagnetic radiation of wavelengths of 10–400 nanometers, shorter than that of visible light, but longer than X-rays. UV radiation is present in sunlight and constitutes about 10% of the total electromagnetic radiation output from the Sun. It is also produced by electric arcs, Cherenkov radiation, and specialized lights, such as mercury-vapor lamps, tanning lamps, and black lights. Extrachromosomal RDNA Circle
Extrachromosomal rDNA circles (aka ERC) are pieces of extrachromosomal circular DNA (eccDNA) derived from ribosomal DNA (rDNA). Initially found in baker's yeast, these self-replicating circles are suggested to contribute to their aging and found in their aged cells. Like ordinary eccDNA, they are created by intra-molecular homologous recombination of the chromosome. The process for intra-molecular homologous recombination is independent of chromosomal replication. The de novo generated circles had exact multiples of tandem copies of 2-kb fragments from cosmid templates. The tandem organization is essential to circle formation. The ribosomal DNA (rDNA) consists of a group of ribosomal RNA encoding genes and related regulatory elements, and is widespread in similar configuration in all domains of life. The ribosomal DNA encodes the non-coding ribosomal RNA, integral structural elements in the assembly of ribosomes, its importance making it the most abundant section of RNA found in cells of eukaryotes. Additionally, these segments include regulatory sections, such as a promoter specific to the RNA polymerase I, as well as both transcribed and non-transcribed spacer segments. A telomere (; ) is a region of repetitive nucleotide sequences associated with specialized proteins at the ends of linear chromosomes (see #Sequences, Sequences).
