Evelyn Maisel Witkin
Evelyn M. Witkin ( Maisel; March 9, 1921 – July 8, 2023) was an American bacterial geneticist at Cold Spring Harbor Laboratory (1944–1955), SUNY Downstate Medical Center (1955–1971), and Rutgers University (1971–1991). Witkin was considered innovative and inspirational as a scientist, teacher and mentor. Her work on DNA damage and DNA repair in bacteria is foundational to our understanding of such processes in living organisms. Her work has direct application to the effects of aging and to the diagnosis and treatment of human diseases such as cancer. Witkin was awarded the National Medal of Science in 2002 for her pioneering work on DNA mutagenesis and DNA repair. In 2015, Witkin received the Albert Lasker Award for Basic Medical Research "for discoveries concerning the DNA-damage response - a fundamental mechanism that protects the genomes of all living organisms." SOS Response
The SOS response is a global transcriptional response to DNA damage in prokaryotes, in which the cell cycle is arrested and DNA repair mechanisms (error-free as well as error-prone) are induced. The regulation of this response is driven by two proteins, RecA and LexA. The RecA protein, stimulated by single-stranded DNA, is involved in the inactivation of the repressor ( LexA) of SOS response genes thereby inducing the response. It is an error-prone repair system that contributes significantly to DNA changes observed in a wide range of bacterial species. Discovery The SOS response was articulated by Evelyn Witkin. Later, by characterizing the phenotypes of mutagenised ''E. coli'', she and post doctoral student Miroslav Radman detailed the SOS response to UV radiation in bacteria. Miroslav Radman (born April 30, 1944) is a Croatian biologist. Biography Radman was born in Split, PR Croatia, Yugoslavia. From 1962–1967 he studied experimental biology, physical chemistry and molecular biology at the University of Zagreb and in 1969 he obtained a doctorate degree in molecular biology at the Free University of Brussels. He spent the next three years at Harvard University as a postdoctoral researcher. From 1973 until 1983 he was Professor of Molecular Biology at the Free University of Brussels and from 1983 until 1998 the Research Director at the French Centre for Scientific Research at the University of Paris 7. Mutation Frequency Decline (''mfd'') is the gene which encodes the protein Mfd (also known as Transcription Repair Coupling Factor, TRCF). Mfd functions in transcription-coupled repair to remove a stalled RNA polymerase that has encountered DNA damage and is unable to continue translocating. The protein is named after the fact that it reduces the chances of suppressor mutations in UV-irradiated cells (or rather, knockout cells show higher rates of such mutations). It does not reduce the chance of every kind of mutation. In fact, it seems to increase the chances of mutation in general, helping to evolve new traits such as antimicrobial resistance. Nucleotide Excision Repair
Nucleotide excision repair is a DNA repair mechanism. DNA damage occurs constantly because of chemicals (e.g. Intercalation (biochemistry), intercalating agents), radiation and other mutagens. Three excision repair pathways exist to repair single stranded DNA damage: Nucleotide excision repair (NER), base excision repair (BER), and DNA mismatch repair (MMR). While the BER pathway can recognize Base excision repair#Lesions processed by BER, specific non-bulky lesions in DNA, it can correct only damaged bases that are removed by Base excision repair#DNA glycosylases, specific glycosylases. Ultraviolet radiation, also known as simply UV, is electromagnetic radiation of wavelengths of 10–400 nanometers, shorter than that of visible light, but longer than X-rays. Barbara McClintock (June 16, 1902 – September 2, 1992) was an American scientist and cytogenetics, cytogeneticist who was awarded the 1983 Nobel Prize in Physiology or Medicine. McClintock received her PhD in botany from Cornell University in 1927. Bernard David Davis (January 7, 1916 – January 14, 1994) was an American biologist who made major contributions in microbial physiology and metabolism. Davis was a prominent figure at Harvard Medical School in microbiology and in national science policy. He was the 1989 recipient of the Selman A. Waksman Award in Microbiology from the National Academy of Sciences. Education Davis was born in Franklin, Massachusetts, where his parents, Jewish immigrants from Lithuania, had settled. Leo Szilard (; ; born Leó Spitz; February 11, 1898 – May 30, 1964) was a Hungarian-born physicist, biologist and inventor who made numerous important discoveries in nuclear physics and the biological sciences. He conceived the nuclear chain reaction in 1933, and patented the idea in 1936.
