Dumas Method
The Dumas method in analytical chemistry is a method for the quantitative determination of nitrogen in chemical substances based on a method first described by Jean-Baptiste Dumas in 1826. The Dumas technique has been automated and instrumentalized, so that it is capable of rapidly measuring the crude protein concentration of food samples. This automatic Dumas technique has replaced the Kjeldahl method as the standard method of analysis for nutritional labelling of protein content of foods (except in high fat content foods where the Kjeldahl method is still preferred due to fire risks). Method The method consists of combusting a sample of known mass to a temperature between 800 and 900 °C in the presence of oxygen. This leads to the release of carbon dioxide, water and nitrogen. In physical and analytical chemistry, colorimetry or colourimetry is a technique used to determine the concentration of colored compounds in solution. A colorimeter is a device used to test the concentration of a solution by measuring its absorbance of a specific wavelength of light (not to be confused with the tristimulus colorimeter used to measure colors in general). To use the colorimeter, different solutions must be made, including a control or reference of known concentration. With a visual colorimeter, for example the Duboscq colorimeter illustrated, the length of the light path through the solutions can be varied while filtered light transmitted through them is compared for a visual match. Bicinchoninic Acid Assay
The bicinchoninic acid assay (BCA assay), also known as the Smith assay, after its inventor, Paul K. Smith at the Pierce Chemical Company, now part of Thermo Fisher Scientific, is a biochemical assay for determining the total concentration of protein in a solution (0.5 μg/mL to 1.5 mg/mL), similar to Lowry protein assay, Bradford protein assay or biuret reagent. The total protein concentration is exhibited by a color change of the sample solution from green to purple in proportion to protein concentration, which can then be measured using colorimetric techniques. Combustion analysis is a method used in both organic chemistry and analytical chemistry to determine the elemental composition (more precisely empirical formula) of a pure organic compound by combusting the sample under conditions where the resulting combustion products can be quantitatively analyzed. Once the number of moles of each combustion product has been determined the empirical formula or a partial empirical formula of the original compound can be calculated. Non-protein Nitrogen
Non-protein nitrogen (or NPN) is a term used in animal nutrition to refer collectively to components such as urea, biuret, and ammonia, which are not proteins but can be converted into proteins by microbes in the ruminant stomach. Due to their lower cost compared to plant and animal proteins, their inclusion in a diet can result in economic gain, but at too high levels cause a depression in growth and possible ammonia toxicity, as microbes convert NPN to ammonia first before using that to make protein. Kjeldahl Method
The Kjeldahl method or Kjeldahl digestion () in analytical chemistry is a method for the quantitative determination of nitrogen contained in organic substances plus the nitrogen contained in the inorganic compounds ammonia and ammonium (NH3/NH4+).
