Propiophenone (shorthand: benzoylethane or BzEt) is an aryl ketone. It is a colorless, sweet-smelling liquid that is insoluble in water, but miscible with organic solvents. It is used in the preparation of other compounds. Production Propiophenone can be prepared by Friedel–Crafts reaction of propanoyl chloride and benzene. It is also prepared commercially by ketonization of benzoic acid and propionic acid over calcium acetate and alumina at 450–550 °C: :C6H5CO2H + CH3CH2CO2H → C6H5C(O)CH2CH3 + CO2 + H2O Ludwig Claisen discovered that Alpha and beta carbon, α-methoxystyrene forms this compound when heated for an hour at 300 °C (65% yield). Diphenidol
Diphenidol is a muscarinic antagonist employed as an antiemetic and as an antivertigo agent. It is not marketed in the United States or Canada. Although the mechanism of action of diphenidol on the vestibular system has not yet been elucidated, it exerts an anticholinergic effect due to interactions with mACh receptors, particularly M1, M2, M3 and M4. Hence, its actions may take place at the vestibular nuclei, where a significant excitatory input is mediated by ACh receptors, and also at the vestibular periphery where mACh receptors are expressed at efferent synapses. A series of selective mACh-receptor antagonists based on the diphenidol molecule has been synthesized, but they have not yet been the subject of clinical trials. Clofedanol
Clofedanol (INN) or chlophedianol ( BAN), sold under the brand name Ninjacof among others, is a centrally acting cough suppressant used in the treatment of dry cough. Clofedanol has local anesthetic, antispasmodic, and antihistamine properties, and may have anticholinergic effects at high doses. Side effects Adverse effects may include irritability, drowsiness, nightmares, vertigo, nausea or vomiting, visual disturbances or hallucinations, and urticaria. There are potential interactions with other anticholinergics, CNS depressants, and alcohol. Pharmacology Unlike many other antitussive drugs such as dextromethorphan, it binds poorly to the sigma-1 receptor. Society and culture Chlophedianol was approved for OTC status in 1987 by the FDA OTC monograph process and its safety and efficacy data are limited.
