David Mangelsdorf
David J. Mangelsdorf is an American biologist and chemist, currently the Alfred J. Gilman Distinguished Chair in Pharmacology, Raymond and Ellen Willie Distinguished Chair in Molecular Neuropharmacology at University of Texas Southwestern Medical Center. In 2008, he was elected to the National Academy of Sciences. He, along with Steven Kliewer, identified the ligands and physiologic functions of a number of orphan nuclear receptors that then discovered two new signaling pathways mediated by the endocrine factors FGF19 and FGF21, which has become a significant accomplishment in the field. He received BS degrees in Biology and Chemistry from Northern Arizona University in 1981 and a PhD in Biochemistry from the University of Arizona in 1987. Fibroblast growth factor 19 is a protein that in humans is encoded by the ''FGF19'' gene. It functions as a hormone, regulating bile acid synthesis, with effects on glucose and lipid metabolism. Reduced synthesis, and blood levels, may be a factor in chronic bile acid diarrhea and in certain metabolic disorders. Fibroblast growth factor 21 is a liver-secreted peptide hormone that in humans is encoded by the ''FGF21'' gene. Together with FGF19 ( FGF15 in rodents) and FGF23, this protein is a member of the endocrine subgroup within the fibroblast growth factor (FGF) family. FGF21 is a potent, extracellularly acting metabolic regulator, whose action was discovered through ''in vitro'' phenotypic screeninig and diet manipulation studies in rodents., unlike canonical growth-stimulating FGFs known to stimulate mitosis, differentiation and angiogenesis in their target tissues, FGF21 exerts its action by activating FGF21 receptors located in the cell membrane of target cells.
