In bacteriology, gram-positive bacteria are bacteria that give a positive result in the Gram stain test, which is traditionally used to quickly classify bacteria into two broad categories according to their type of cell wall. Gram-positive bacteria take up the crystal violet stain used in the test, and then appear to be purple-coloured when seen through an optical microscope. This is because the thick peptidoglycan layer in the bacterial cell wall retains the stain after it is washed away from the rest of the sample, in the decolorization stage of the test. Conversely, gram-negative bacteria cannot retain the violet stain after the decolorization step; alcohol used in this stage degrades the outer membrane of gram-negative cells, making the cell wall more porous and incapable of retaining the crystal violet stain. Spent mushroom compost is the residual compost waste generated by the mushroom production industry. Background It is readily available (bagged, at nursery suppliers), and its formulation generally consists of a combination of wheat straw, dried blood, horse manure and ground chalk, composted together. It is an excellent source of humus, although much of its nitrogen content will have been used up by the composting and growing mushrooms. It remains, however, a good source of general nutrients (0.7% N, 0.3% P, 0.3% K plus a full range of trace elements), as well as a useful soil conditioner. However, due to its chalk content, it may be alkaline, and should not be used on acid-loving plants, nor should it be applied too frequently, as it will overly raise the soil's pH levels.
