Glycobiology
Defined in the narrowest sense, glycobiology is the study of the structure, biosynthesis, and biology of saccharides ( sugar chains or glycans) that are widely distributed in nature. Sugars or saccharides are essential components of all living things and aspects of the various roles they play in biology are researched in various medical, biochemical and biotechnological fields. History According to ''Oxford English Dictionary'' the specific term ''glycobiology'' was coined in 1988 by Prof. Raymond Dwek to recognize the coming together of the traditional disciplines of carbohydrate chemistry and biochemistry. This coming together was as a result of a much greater understanding of the cellular and molecular biology of glycans. Raymond Dwek
Raymond Allen Dwek CBE FRS FRSC (born 10 November 1941) is a scientist at the University of Oxford and co-founder of the biotechnology company Oxford GlycoSciences Ltd., Biography Dwek was educated at Carmel College, and the University of Manchester, where he studied chemistry (1960–64). He then went to Oxford University and Lincoln College, Oxford, where he completed his DPhil in physical chemistry in 1966. He became Professor of Glycobiology in 1988 in the Department of Biochemistry. He is an emeritus fellow of Exeter College, Oxford and co-director of the Oxford Glycobiology Institute, which he founded in 1991. From 2000 to 2006 he was also head of the Department of Biochemistry. Rosalind Kornfeld
Rosalind Hauk Kornfeld (1935–2007) was a scientist at Washington University in St. Louis known for her research determining the structure and formation of oligosaccharides. The Society of Glycobiology annually awards a lifetime achievement award in her honor. Education and career Rosalind Kornfeld Hauk was born in Dallas, Texas and then grew up in Chevy Chase, Maryland. She earned a bachelor of science degree from George Washington University in 1957. She went on to receive her Ph.D. degree in 1961 working on enzymes in rabbit muscles. For a brief period she stayed at George Washington University as a postdoctoral investigator before moving to the National Institutes of Health as a fellow working with Victor Ginsburg. She moved back to St. Louis in 1965 when she accepted a position at Washington University in St. Louis. Glycan-Protein interactions represent a class of biomolecular interactions that occur between free or protein-bound glycans and their cognate binding partners. Intramolecular glycan-protein (protein-glycan) interactions occur between glycans and proteins that they are covalently attached to. Together with protein-protein interactions, they form a mechanistic basis for many essential cell processes, especially for cell-cell interactions and host-cell interactions. For instance, SARS-CoV-2, the causative agent of COVID-19, employs its extensively glycosylated spike (S) protein to bind to the ACE2 receptor, allowing it to enter host cells. Glycan
The terms glycans and polysaccharides are defined by IUPAC as synonyms meaning "compounds consisting of a large number of monosaccharides linked glycosidically". However, in practice the term glycan may also be used to refer to the carbohydrate portion of a glycoconjugate, such as a glycoprotein, glycolipid, or a proteoglycan, even if the carbohydrate is only an oligosaccharide. Glycans usually consist solely of O-glycosidic linkages of monosaccharides. For example, cellulose is a glycan (or, to be more specific, a glucan) composed of β-1,4-linked D-glucose, and chitin is a glycan composed of β-1,4-linked ''N''-acetyl-D-glucosamine. Glycomics
Glycomics is the comprehensive study of glycomes (the entire complement of sugars, whether free or present in more complex molecules of an organism), including genetic, physiologic, pathologic, and other aspects. Glycomics "is the systematic study of all glycan structures of a given cell type or organism" and is a subset of glycobiology. The term glycomics is derived from the chemical prefix for sweetness or a sugar, "glyco-", and was formed to follow the '' omics'' naming convention established by genomics (which deals with genes) and proteomics (which deals with proteins). Glycoproteins are proteins which contain oligosaccharide chains covalently attached to amino acid side-chains. The carbohydrate is attached to the protein in a cotranslational or posttranslational modification. This process is known as glycosylation. Secreted extracellular proteins are often glycosylated. In proteins that have segments extending extracellularly, the extracellular segments are also often glycosylated. Glycoproteins are also often important integral membrane proteins, where they play a role in cell–cell interactions. It is important to distinguish endoplasmic reticulum-based glycosylation of the secretory system from reversible cytosolic-nuclear glycosylation. Erythropoietin
Erythropoietin (; EPO), also known as erythropoetin, haematopoietin, or haemopoietin, is a glycoprotein cytokine secreted mainly by the kidneys in response to cellular hypoxia; it stimulates red blood cell production (erythropoiesis) in the bone marrow. Low levels of EPO (around 10 mU/mL) are constantly secreted in sufficient quantities to compensate for normal red blood cell turnover. Nucleotides are organic molecules consisting of a nucleoside and a phosphate.
