Gracilicutes
Gracilicutes (Latin: ''gracilis'', slender, and ''cutis'', skin, referring to the cell wall) is a clade in bacterial phylogeny. Traditionally gram staining results were most commonly used as a classification tool, consequently until the advent of molecular phylogeny, the Kingdom Monera (as the domains Bacteria and Archaea were known then) was divided into four phyla, * Gracilicutes (gram-negative, it is split in many groups, but some authors still use it in a narrower sense) * Firmacutes ic(gram-positive, subsequently corrected to Firmicutes, today it excludes the Actinomycetota) * Mollicutes (gram variable, later renamed Tenericutes and now Mycoplasmatota, e.g. ''Mycoplasma'') * Mendosicutes (uneven gram stain, "methanogenic bacteria" now known as methanogens and classed as Archaea) This classification system was abandoned in favour of the three-domain system based on molecular phylogeny started by C. Hydrobacteria
Hydrobacteria is a taxon containing approximately one-third of prokaryote species, mostly gram-negative bacteria and their relatives. It was found to be the closest relative of an even larger group of Bacteria, Terrabacteria, which are mostly gram positive bacteria. The name Hydrobacteria (hydro = "water") refers to the moist environment inferred for the common ancestor of those species. In contrast, species of Terrabacteria possess adaptations for life on land. The content of Hydrobacteria has grown to include these superphyla and phyla: Acidobacteriota, Aquificota, Bdellovibrionota, Campylobacterota, Deferribacterota, Dependentiae, Desulfobacterota, Desulfuromonadota, Elusimicrobiota, FCB superphylum, Myxococcota, Nitrospirota, Proteobacteria, PVC superphylum, and Spirochaetota. Monera
Monera (/məˈnɪərə/) (Greek - μονήρης (monḗrēs), "single", "solitary") is a biological kingdom that is made up of prokaryotes. As such, it is composed of single-celled organisms that lack a nucleus. The taxon Monera was first proposed as a phylum by Ernst Haeckel in 1866. Subsequently, the phylum was elevated to the rank of kingdom in 1925 by Édouard Chatton. The last commonly accepted mega-classification with the taxon Monera was the five-kingdom classification system was established by Robert Whittaker in 1969. Gram-positive Bacteria
In bacteriology, gram-positive bacteria are bacteria that give a positive result in the Gram stain test, which is traditionally used to quickly classify bacteria into two broad categories according to their type of cell wall. Gram-positive bacteria take up the crystal violet stain used in the test, and then appear to be purple-coloured when seen through an optical microscope. This is because the thick peptidoglycan layer in the bacterial cell wall retains the stain after it is washed away from the rest of the sample, in the decolorization stage of the test. Conversely, gram-negative bacteria cannot retain the violet stain after the decolorization step; alcohol used in this stage degrades the outer membrane of gram-negative cells, making the cell wall more porous and incapable of retaining the crystal violet stain. Mollicutes
Mollicutes is a class of bacteria distinguished by the absence of a cell wall. The word "Mollicutes" is derived from the Latin ''mollis'' (meaning "soft" or "pliable"), and ''cutis'' (meaning "skin"). Individuals are very small, typically only 0.2–0.3 μm (200-300 nm) in size and have a very small genome size. They vary in form, although most have sterols that make the cell membrane somewhat more rigid. Many are able to move about through gliding, but members of the genus '' Spiroplasma'' are helical and move by twisting. The best-known genus in the Mollicutes is ''Mycoplasma''. Planctobacteria
The PVC superphylum is a superphylum of bacteria named after its three important members, Planctomycetota, Verrucomicrobiota, and Chlamydiota. Cavalier-Smith postulated that the PVC bacteria probably lost or reduced their peptidoglycan cell wall twice. It has been hypothesised that a member of the PVC clade might have been the host cell in the endosymbiotic event that gave rise to the first proto-eukaryotic cell. Proteobacteria
Pseudomonadota (synonym Proteobacteria) is a major phylum of Gram-negative bacteria. The renaming of phyla in 2021 remains controversial among microbiologists, many of whom continue to use the earlier names of long standing in the literature. ''Mycoplasma'' is a genus of bacteria that, like the other members of the class '' Mollicutes'', lack a cell wall around their cell membranes. Peptidoglycan ( murein) is absent. Capsule (microbiology)
The bacteria capsule is a large structure common to many bacteria. It is a polysaccharide layer that lies outside the cell envelope, and is thus deemed part of the outer envelope of a bacterial cell. It is a well-organized layer, not easily washed off, and it can be the cause of various diseases. The capsule—which can be found in both gram negative and gram-positive bacteria—is different from the second lipid membrane – bacterial outer membrane, which contains lipopolysaccharides and lipoproteins and is found only in gram-negative bacteria. When the amorphous viscid secretion (that makes up the capsule) diffuses into the surrounding medium and remains as a loose undemarcated secretion, it is known as a slime layer.
