Frizzled
Frizzled is a family of atypical G protein-coupled receptor, G protein-coupled receptors that serve as receptors in the Wnt signaling pathway and other signaling pathways. When activated, Frizzled leads to activation of Dishevelled in the cytosol. Species distribution Frizzled proteins and the genes that encode them have been identified in an array of animals, from sponges to humans. Function Frizzled proteins also play key roles in governing cell polarity, embryonic development, formation of neural synapses, cell proliferation, and many other processes in developing and adult organisms. These processes occur as a result of one of three signaling pathways. FZD1
Frizzled-1 (Fz-1) is a protein that in humans is encoded by the ''FZD1'' gene. Function Members of the 'frizzled' gene family encode G protein–coupled receptor, 7-transmembrane domain proteins that are receptors for Wnt signaling proteins. FZD4
Frizzled-4 (Fz-4) is a protein that in humans is encoded by the ''FZD4'' gene. Fz-4 has also been designated as CD344 (cluster of differentiation 344). Function This gene is a member of the frizzled gene family. Members of this family encode seven-transmembrane domain proteins that are receptors for the Wingless type MMTV integration site family of signaling proteins. Frizzled-4 is the only representative of frizzled family members that binds strongly an additional ligand Norrin that is functionally similar but structurally different from Wingless type proteins. FZD4 signaling induced by Norrin regulates vascular development of vertebrate retina and controls important blood vessels in the ear. FZD6
Frizzled-6 (Fz-6) is a protein that in humans is encoded by the ''FZD6'' gene. Members of the 'frizzled' gene family encode 7-transmembrane domain proteins that are receptors for WNT signaling proteins. The FZD6 protein contains a signal peptide, a cysteine-rich domain in the N-terminal extracellular region, and 7 transmembrane domains. FZD3
Frizzled-3 (Fz-3) is a protein that in humans is encoded by the ''FZD3'' gene. Function This gene is a member of the frizzled gene family. Members of this family encode seven-transmembrane domain proteins that are receptors for the Wingless type MMTV integration site family of signaling proteins. Most frizzled receptors are coupled to the beta-catenin canonical signaling pathway. It may play a role in mammalian hair follicle development. The function of this gene is largely derived from mouse studies. Fzd3 in the mouse functions through planar cell polarity signaling instead of the canonical Wnt/beta-catenin pathway. Vantictumab
Vantictumab is a human IgG2 monoclonal antibody designed for the treatment of cancer. This drug was developed by OncoMed Pharmaceuticals Inc. in partnership with Bayer. Wnt Signaling Pathway
In cellular biology, the Wnt signaling pathways are a group of signal transduction pathways which begin with proteins that pass signals into a cell through cell surface receptors. The name Wnt, pronounced "wint", is a portmanteau created from the names Wingless and Int-1. Wnt signaling pathways use either nearby cell-cell communication (paracrine) or same-cell communication (autocrine). They are highly evolutionarily conserved in animals, which means they are similar across animal species from fruit flies to humans. Three Wnt signaling pathways have been characterized: the canonical Wnt pathway, the noncanonical planar cell polarity pathway, and the noncanonical Wnt/calcium pathway.
