Philadelphia Chromosome
The Philadelphia chromosome or Philadelphia translocation (Ph) is an abnormal version of chromosome 22 where a part of the ''ABL (gene), Abelson murine leukemia'' 1 (''ABL1'') gene on chromosome 9 breaks off and attaches to the ''BCR (gene), breakpoint cluster region'' (''BCR'') gene in chromosome 22. The balanced reciprocal Translocation (genetics), translocation between the long arms of 9 and 22 chromosomes [t (9; 22) (q34; q11)] results in the fusion gene ''BCR::ABL1''. The Oncogene, oncogenic protein with persistently enhanced tyrosine kinase (TK) activity transcribed by the ''BCR''::''ABL1'' fusion gene can lead to rapid, uncontrolled growth of immature white blood cells that accumulates in the blood and bone marrow. Mixed-phenotype Acute Leukemia
Mixed-phenotype acute leukemia (MPAL) is a group of blood cancers (leukemia) which have combined features of myeloid and lymphoid cancers. It is a rare disease, constituting about 2–5% of all leukemia cases. It mostly involve myeloid with either of T lymphocyte or B lymphocyte progenitors, but in rare cases all the three cell lineages. Knowledge on the cause, clinical features and cellular mechanism is poor, making the treatment and management (prognosis) difficult. The name "mixed-phenotype acute leukemia" was adopted by the World Health Organization in 2008 to include leukemias of ambiguous lineage, acute undifferentiated leukemias and natural killer lymphoblastic leukemias. Retrovirus
A retrovirus is a type of virus that inserts a DNA copy of its RNA genome into the DNA of a host cell that it invades, thus changing the genome of that cell. After invading a host cell's cytoplasm, the virus uses its own reverse transcriptase enzyme to produce DNA from its RNA genome, the reverse of the usual pattern, thus ''retro'' (backward). The new DNA is then retroviral integration, incorporated into the host cell genome by an integrase enzyme, at which point the retroviral DNA is referred to as a provirus. Abelson Murine Leukemia Virus
The Abelson murine leukemia virus (Ab-MLV or A-MuLV) is a retrovirus (Class VI) used to induce malignant transformation of murine lymphoid cells. As a retrovirus, it has a single-stranded, positive sense RNA genome which replicates via a DNA intermediate mediated by a reverse transcriptase. The Abelson murine leukemia virus is named for the American pediatrician Herbert T. Abelson, who together with Louise S Rabstein, first described and isolated it. A-MuLV causes a rapidly progressive lymphosarcoma known as Abelson disease in mice, which is a type of leukemia that does not involve the thymus. However, this is only possible when the host cell is co-infected with a helper virus which provides functions it needs to be able to replicate which it does not code for in its own genome such as a reverse transcriptase and some major structural proteins.
